In 2012 the World Health Organisation reported about 250 million preschool children were affected by vitamin A deficiency (VAD), and that providing those children with vitamin A could prevent about a third of all under-five deaths, which amounts to up to 2.7 million children that could be saved from dying unnecessarily [1]. It is known that millions of people each year are dying from VAD and many are being affected by its serve force. Rice containing provitamin A could substantially reduce the problems of this dreadful deficiency which “…can only be achieved using genetic engineering because there is no provitamin A in the rice seeds, even though it is present in the leaves” [1]. Golden Rice technology is founded on the simple principle that “…by adding only two genes, a plant phytoene synthase (psy) and a bacterial phytoene desaturase (crt I), the pathway is turned back on and β-carotene consequently accumulates in the grain” [1].

On the other hand, the non-carotenogenic starchy rice endosperm, is very low in lipid and lacks any means for carotenoid deposition. It was also doubtful whether Golden Rice would have the essential predecessors for carotene biosynthesis current and obtainable in the grains, with the belief that the whole, multi-step carotenoid biosynthetic pathway was entirely missing from the endosperm. This is why such a long research phase preceded after the proof of the concept of Golden Rice.
